Vumerity (diroximel fumarate) and Mayzent (siponimod) are used to treat relapsing forms of multiple sclerosis (MS), to include clinically isolated syndrome, relapsing-remitting disease, and active secondary progressive disease, in adults.

Vumerity and Mayzent belong to different drug classes. Vumerity is an immunomodulator and Mayzent is a sphingosine 1-phosphate receptor modulator.

Side effects of Vumerity and Mayzent that are similar include diarrhea and nausea.

Side effects of Vumerity that are different from Mayzent include flushing, abdominal pain, vomiting, itching, rash, albumin in the urine, redness, and indigestion/heartburn.

Side effects of Mayzent that are different from Vumerity include headache, high blood pressure (hypertension), increased transaminase, falls, dizziness, slow heart rate, swelling of extremities, and pain in extremities.

Vumerity may interact with dimethyl fumarate.

Mayzent may interact with "live" vaccines, cancer medications, immune-modulating drugs, immunosuppressive therapies, anti-arrhythmics, QT prolonging drugs, calcium channel blockers, other drugs that may decrease heart rate, beta-blockers, fluconazole, rifampin, carbamazepine, modafinil, and efavirenz.

Common side effects of Vumerity include:

• flushing,

• diarrhea,
• nausea,
• vomiting,
• itching,
• rash,
• albumin in the urine,
• redness, and
• indigestion/heartburn

Common side effects of Mayzent include:

• headache,
• high blood pressure (hypertension),

• pain in extremities

Mayzent (siponimod) is a sphingosine 1-phosphate receptor modulator indicated for the treatment of relapsing forms of multiple sclerosis (MS), to include clinically isolated syndrome, relapsing-remitting disease, and active secondary progressive disease, in adults.

Vumerity may interact with other medicines such as:

• dimethyl fumarate

The starting dose of Vumerity is 231 mg twice a day, orally, for 7 days. The maintenance dose of Vumerity after 7 days is 462 mg (administered as two 231 mg capsules) twice a day, orally.

Initiate Mayzent with a 5-day titration. The recommended maintenance dosage of Mayzent is 2 mg.
