Mayzent (siponimod) is a sphingosine 1-phosphate receptor modulator used to treat relapsing forms of multiple sclerosis (MS), to include clinically isolated syndrome, relapsing-remitting disease, and active secondary progressive disease, in adults.

Tysabri (natalizumab) is a monoclonal antibody used in to treat relapsing forms of multiple sclerosis. Tysabri is also used to treat moderate to severe Crohn's disease in adults. Tysabri is usually given after other Crohn's disease medications have been tried without successful treatment of this condition.

Side effects of Mayzent and Tysabri that are similar include headache, pain and swelling in extremities, and diarrhea.

Side effects of Mayzent that are different from Tysabri include high blood pressure (hypertension), increased liver transaminase, falls, nausea, dizziness, and slow heart rate.

Side effects of Tysabri that are different from Mayzent include tired feeling, joint or muscle pain, redness or irritation at the injection site, changes in menstrual cycle, stomach pain, skin rash, depression, painful menstrual cramps, and cold symptoms such as stuffy nose, sneezing, or sore throat.

Both Mayzent and Tysabri may interact with cancer medications, immune-modulating drugs, and immunosuppressive therapies.

Mayzent may also interact with “live” vaccines, anti-arrhythmics, QT prolonging drugs, calcium channel blockers, other drugs that may decrease heart rate, beta-blockers, fluconazole, rifampin, carbamazepine, modafinil, and efavirenz.

Tysabri may also interact with other medicines, especially those that may affect the immune system such as interferon, cyclosporine, sirolimus, tacrolimus, basiliximab, efalizumab, muromonab-CD3, mycophenolate mofetil, azathioprine, leflunomide, etanercept, or chemotherapy or radiation treatments.

Common side effects of Mayzent include:

• headache,
• high blood pressure (hypertension),
• increased transaminase,
• falls,
• swelling of extremities,
• nausea,
• dizziness,
• diarrhea,
• slow heart rate, and
• pain in extremities

Common side effects of Tysabri include:

• headache,
• tired feeling,
• joint or muscle pain,
• redness or irritation at the injection site,
• swelling hands/feet/ankles,
• changes in menstrual cycle,
• stomach pain,
• diarrhea,
• skin rash,
• depression,
• painful menstrual cramps, or
• cold symptoms such as stuffy nose, sneezing, or sore throat.

Tell your doctor if you have side effects while Tysabri is being given or shortly after your treatment is finished (infusion reaction) including:

• chills,
• fever,
• flushing,
• nausea,
• dizziness, and
• chest pain.

Initiate Mayzent with a 5-day titration. The recommended maintenance dosage of Mayzent is 2 mg.

The recommended dose of Tysabri for multiple sclerosis or Crohn's disease is 300 mg intravenous infusion over one hour every four weeks.
